Copying
If you copy a folder (and its content) and make a change in one of the duplicate folders or even the file, will it reflect in the original folder or file?

To answer your question, no, the changes in one of the duplicate file (assuming that you copied that file or folder where that file is located) will not reflect on the original file.
Rather than creating a duplicate copy of files from a folder to another, you can Bookmark the original file to achieve your desired goal here and copy that bookmark in multiple folders in your Box.
The user/s using the bookmark would have to have access to the actual folder (where the bookmark points to) as well.
